ABSTRACT

Biosecurity, cleaning and disinfection of swine and poultry facilities are fundamental for the reduction of pathogenic microorganisms of importance for public and animal health. The objective of this work was to compare the levels of active ingredient described on the label and the real levels detected in high-performance liquid chromatography (HPLC) of two disinfectants., then evaluate the antimicrobial activity since, following the Germicidal Sanitizing Action and Disinfectant Detergent (Official Method AOAC 960.09) in four different dilutions with the presence of 3% organic matter during 15 min of contact, against Salmonella Heidelberg and Salmonella Typhimurium (ST). The product "A" presents active levels of agreement according to the label. The content of quantified assets for product "B" was lower than that recorded on the label. The disinfectant "A" was effective in microbiological evaluation while the disinfectant "B" had microbiocidal activity compromised by the deficit of assets.

ABSTRACT

Salmonella is a major cause of foodborne illness worldwide, and poultry and its derived products are the most common food products associated with salmonellosis outbreaks. Some countries, including Brazil, have experienced an increased prevalence of Salmonella Heidelberg among their poultry flocks. Some isolates have also presented high resistance to antimicrobial agents and persist in the poultry farm environment. This study aimed to compare the susceptibility of S. Heidelberg strains isolated in 2006 with those isolated in 2016 against disinfectants and antimicrobial agents. The results showed that all the strains were highly susceptible to sodium hypochlorite, regardless of the conditions and year of isolation. Resistance to benzalkonium chloride varied according to the conditions applied, but not to the year of isolation. Increased antimicrobial resistance from 2006-2016 was observed only for tetracycline. The results suggest that the antimicrobial and disinfectant resistance of S. Heidelberg did not increase for ten years (2006-2016). However, further analysis should include a larger number of S. Heidelberg isolates from poultry origin and additional antimicrobial agents for more precise conclusions about the increasing in antimicrobial resistance in the last years.(AU)


Salmonella é uma das principais causas das doenças transmitidas por alimento em todo o mundo, e a carne de frango e produtos derivados são os principais alimentos associados com surtos de salmonelose em humanos. Alguns países, incluindo o Brasil, têm observado um aumento da ocorrência de Salmonella Heidelberg nas suas granjas avícolas. Além disto, alguns isolados têm apresentado alta resistência aos antimicrobianos e têm persistido no ambiente de produção avícola. Neste contexto, o objetivo deste estudo foi comparar a susceptibilidade de cepas de S. Heidelberg isoladas em 2006 com aquelas isoladas em 2016 contra desinfetantes e agentes antimicrobianos. Os resultados demonstraram que as cepas foram altamente resistentes a hipoclorito de sódio, independentemente das condições e do ano de isolamento. A resistência ao cloreto de benzalcônio variou de acordo com as condições testadas, mas não com o ano de isolamento. Um aumento da resistência aos antimicrobianos de 2006 a 2016 foi observado apenas para tetraciclina. Os resultados sugerem que a resistência aos desinfetantes e aos antimicrobianos não aumentou em um período de dez anos (2006-2016). Entretanto, novas análises devem incluir um número maior de cepas de S. Heidelberg isoladas de fontes avícolas e outros agentes antimicrobianos para uma conclusão mais precisa sobre o aumento da resistência antimicrobiana nos últimos anos.(AU)

ABSTRACT

O objetivo deste estudo foi de avaliar a atividade antifúngica de uma solução à base de glucoprotamina (GLU) e de óleo essencial (OE) de Cymbopogon citratus. Candida tropicalis, Claviceps lusitaniae, Meyerozyma (Pichia) guilliermondii e Candida parapsilosis foram isoladas das superfícies de um hospital público de Pelotas, RS. A atividade antifúngica da GLU foi analisada nas concentrações de 0,25% a 0,0005%, e para o OE, obtido por Clevenger, foi utilizado nas concentrações de 17.500 μg/mL a 0,034 μg/mL. A análise cromatográfica foi efetuada por meio de CG/MS. A atividade anti-Candida foi analisada pela técnica de Microdiluição em Caldo para determinar a Concentração Inibitória Mínima (CIM) e a Concentração Fungicida Mínima (CFM). A inibição pela GLU ocorreu em todas as concentrações testadas, enquanto o OE apresentou resultado de CIM que oscilou de 2846 a 5450 μg/mL, e CFM de 2853 a 5330 μg/mL. Alfa–citral (37,10%), beta–citral (32,77% e beta-mirceno (18,75%) foram considerados como os compostos majoritários do OE. Os dados obtidos neste trabalho destacam a atividade antifúngica da solução à base de GLU e do OE de C. citratus na eliminação dos isolados de Candida spp ambientais.


This study aimed at evaluating the antifungal activity of glucoprotamine solution (GLU) and the essential oil from Cymbopogon citratus (EO). Candida tropicalis, Claviceps lusitaniae, Meyerozyma (Pichia) guilliermondii and Candida parapsilosis were isolated from the samples, which were collected from the surfaces of a public hospital located in Pelotas, RS. The antifungal activity of GLU was detected at the concentrations from 0.25% to 0.0005%, while the EO, by Clevenger, was used at concentrations from 17.500 μg mL to 0.034 μg/mL. Chemical analysis was determined by GC/MS. The anti-Candida activity was performed by using Broth Microdilution technique to determine the Minimal Fungicidal Concentration (MFC) and the Minimal Inhibitory Concentration (MIC). The inhibition by GLU occurred at all of the tested concentrations, whereas the EO showed MIC ranging from 2.846 to 5.450 μ/mL and the CFM from 2.853 to 5.330 μg/mL. Alfa–citral (37.10%), beta–citral (32.77%) and beta-mirceno (18.75%) were the majority compounds in the EO. The data obtained in the present study highlight the effectiveness of the solution based on GLU and EO for eliminating the environmental isolates of Candida spp.

ABSTRACT

No Brasil, do ano de 2000 a 2017, ocorreram 12.503 surtos de origem alimentar e os locais que sobressaíram como os mais frequentes na ocorrência destes surtos foram restaurantes e residências tendo como uma das causas principais, as superfícies de equipamentos, utensílios e objetos mal higienizados. O produto mais usado para realizar a higiene é o hipoclorito de sódio (água sanitária), sendo este utilizado tanto para serviços de alimentação como para uso residencial. O hipoclorito é comercializado industrialmente e este pode ser fabricado de forma impropria não correspondendo ao percentual de cloro contido em sua rotulagem. O objetivo deste estudo foi analisar o teor de cloro das marcas comerciais de águas sanitárias industrializadas comercializadas na cidade de Maceió/ AL. A metodologia utilizada para determinação de Cloro Ativo foi o método volumétrico, em solução de hipoclorito de sódio. Duas marcas comerciais apresentaram valores menores que 2% de cloro, sete com valores acima e três com valores entre 2 e 2,5%, valores estes preconizados pela legislação. Os resultados mostram que a fabricação deste produto precisa de fiscalização por órgãos competentes para que o teor de cloro esteja dentro do padrão da Legislação. Com essa inadequação a população será prejudicada, pois estará comprando um produto adulterado.


In Brazil, from the year 2000 to 2017, 12,503 foodborne outbreaks have occurred, and the other important local as the more frequent occurrence of these outbreaks was restaurants and residences having as one of the main causes, the surfaces of equipment, utensils and objects poorly sanitized. The product used to perform hygiene is sodium hypochlorite (bleach), which is used for both food service as for residential use. The hypochlorite is sold industrially and this can be manufactured in a manner unbecoming not corresponding to the percentage of chlorine contained in your labelling. The aim of this study was to analyze the chlorine content of the trademarks of sanitary water marketed in the industrialised city of Maceió/AL. The methodology used for the determination of active chlorine, was using the volumetric method, solution of sodium hypochlorite. Two trademarks presented values less than 2% chlorine, seven with values above and 3 with values between 2 and 2.5%, these values provided by the legislation. The results show that the manufacture of this product needs supervision by competent bodies so that the chlorine content is within the pattern of legislation. Whit this inadequacy to population will suffer, as will be buying and adulterated product.

ABSTRACT

Objetivou-se determinar atividade antisséptica do óleo essencial de Lippia origanoides na presença de leite bovino. A composição química do óleo essencial de alecrim pimenta foi determinada por cromatografia gasosa acoplada à espectrometria de massas (CG-EM), sendo detectados 53 compostos, dos quais 16 foram identificados (>0,1% área total). O carvacrol (32,7%), p-cimeno (23%), timilmetil éter, cariofileno (7,98%) e o γ-terpineno (5,40%) foram os componentes mais abundantes. A concentração inibitória mínima para Staphylococcus aureus ATCC 25923 e E. coli ATCC 8739 foi de 60µL/mL, enquanto para Salmonella Choleraesuis ATCC 35640 foi de 90µL/mL. A concentração bactericida mínima foi de 120µL/mL para as três bactérias. Avaliou-se a atividade antisséptica do óleo essencial na concentração de 120µL/mL na presença de leite bovino sobre as mesmas cepas bacterianas. O óleo em estudo apresentou efeito inibitório do crescimento das cepas em diferentes tempos de ação (p<0,05). S. aureus apresentou maiores índices de inibição após 5 min de contato e Escherichia coli e Salmonella Choleraesuis após 15 min. O óleo essencial de alecrim-pimenta revelou ser um potencial antimicrobiano natural, mesmo na presença de matéria orgânica constituída de uma matriz nutricional complexa que é o leite bovino.(AU)


This study aimed to determine the antiseptic activity of the essential oil of Lippia origanoides in the presence of bovine milk. The essential oil chemical composition was determined by gas chromatography-mass spectrometry (CG EM) 53 compounds were detected, of which 16 were identified (>0.1% total area). The carvacrol (32.7%), p-cymene (23%), thymil methyl ether (10.03%), caryophyllene (7.98 %) and γ-terpinene (5.40%) were the most abundant components. The minimum inhibitory concentration for Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 8739 was 60uL/mL, as for this Salmonella Choleraesuis ATCC 35640 was 90uL/mL. The minimum bactericidal concentration was 120µL/mL for all three bacteria. We evaluated the antiseptic activity of the essential oil in the concentration of 120µL/mL in the presence of bovine milk for the same bacterial strains. The oil under study It showed an inhibitory effect of growth of the strains in different action times (p <0.05). S. aureus showed higher inhibition rates after 5 min of contact and E. coli and Salmonella Choleraesuis after 15 min. The essential oil Lippia origanoides proved to be a natural antimicrobial potential even in the presence of organic matter consists of a complex nutritional matrix is bovine milk.(AU)

Em vista de um estudo desenvolvido no Setor de Saneantes do Instituto Nacional de Controle deQualidade em Saúde da Fundação Oswaldo Cruz, referente à avaliação da atividade antimicrobiana dedesinfetantes, no presente trabalho foram analisados produtos desinfetantes de uso geral disponíveis nomercado brasileiro. Os desinfetantes foram coletados aleatoriamente, dos quais três (produtos A, B e C),apresentaram contaminação microbiana. Para efetuar o isolamento e a identificação dos microrganismoscontaminantes foram utilizados o aparelho Vitek® 2, a amplificação e o sequenciamento do gene rRNA16S. A análise realizada por meio de Vitek® 2 revelou a presença das bactérias Serratia marcescens eAchromobacter xylosoxidans, respectivamente, nos produtos A e B. No produto C foram detectadasAeromonas salmonicida pelo Vitek® 2 e Burkholderia lata pela técnica de amplificação da reação em cadeiada polimerase.

Os ambientes hospitalares podem albergar micro-organismos patogênicos e oportunistas, capazes de infectar, particu- larmente, os indivíduos imunossuprimidos. O processo de limpeza e desinfecção assume importância capital para o controle de tais doenças. O presente trabalho avaliou a suscetibilidade de leveduras isoladas de superfícies de ambiente hospitalar veterinário frente a quatro desinfetantes/antissépticos comumente utilizados na rotina de desinfecção. O teste de microdiluição em caldo foi realizado com 24 isolados leveduriformes frente ao hipoclorito de sódio 4%, cloreto de benzalcônico 2%, clorexidina-cetrimida 6,6% e derivado de cloro-fenol 3%. A clorexidina-cetrimida, cloreto de ben- zalcônio e derivado de cloro-fenol foram eficazes em todos os isolados com concentração inibitória mínima e concen- tração fungicida inferiores à concentração recomendada pelo fabricante. Por outro lado, a ação fungicida do hipoclorito de sódio em 79,1% dos isolados testados foi obtida na concentração recomendada pelo fabricante, com desempenho inferior aos demais desinfetantes avaliados.


Hospital environments can have pathogens and opportunistic pathogens, important for immunocompromising individuals. The process of cleaning and disinfection constitutes an important measure for the control of these diseases. This study evaluated the sensitivity of yeasts isolated from surfaces of nosocomial veterinary environment to four disinfectants/antiseptics used in the routine of disinfection. The test of broth microdilution was carried in 24isolates of yeasts against 4% sodium hypochlorite, 2% benzalkonium chloride, 6.6% chlorexidine-cetrimide and 3% chloro-phenol derivate. Chlorexidine-cetrimide, benzalkonium chloride and chloro-phenol derivate were efficient in all isolates with minimal inhibitory concentration and minimal fungicide concentration lower than the concentra- tion recommended by manufacturer. By other hand, fungicide action of the sodium hypochlorite in the concentration recommended by the manufacturer was verified in 79.1% of the isolated tested, with the lower performance of than the others evaluated disinfectants.

ABSTRACT

The purpose of this study was to investigate and compare the efficacy of various disinfectants on planktonic cells and biofilm cells of Listeria monocytogenes, Staphylococcus aureus and Escherichia coli. Numbers of viable biofilm cells decreased after treatment with all tested disinfectants (iodine, biguanide, quaternary ammonium compounds, peracetic acid and sodium hypochlorite). Sodium hypochlorite was the most effective disinfectant against biofilm cells, while biguanide was the least effective. Scanning electron microscopy observations revealed that cells adhered on stainless steel surface after treatment with the disinfectants. No viable planktonic cells were observed after treatment with the same disinfectants. Based on our findings, we concluded that biofilm cells might be more resistant to disinfectants than plancktonic cells.

Objetivo. Evaluar el método de dilución neutralización propuesto en la Norma Técnica Colombiana 5473 de 2007, mediante la utilización de un desinfectante en gel a base de alcohol. Materiales y métodos. El ensayo se efectuó utilizando Pseudomonas aeruginosa ATCC 15442, Staphylococcus aureus ATCC 6538 y Enterococcus hirae ATCC 10541, como microorganismos de ensayo. Las temperaturas del estudio fueron 20±1°C como temperatura obligatoria y 36±1°C y se emplearon cuatro tiempos de contacto entre el desinfectante y los microorganismos evaluados (0, 2, 5 y 10 minutos). El método fue realizado bajo condiciones limpias (0,3 g/L de albumina de suero bovino) y sucias (3g/L de albumina de suero bovino y 3g/L de eritrocitos de oveja). Resultados. La implementación del método arrojó resultados precisos en cada una de las seis repeticiones realizadas en el ensayo. Los resultados obtenidos demostraron una reducción logarítmica superior a cinco, evidenciando la actividad bactericida ejercida por el desinfectante frente a los microorganismos control. El establecimiento de las condiciones experimentales y de la metodología demostró no incidir negativamente en el crecimiento de cada una de las cepas. Igualmente, el neutralizante utilizado no inhibió el desarrollo de los organismos de ensayo. Conclusiones. Se verificó el método mediante el cumplimiento de los límites establecidos por la norma. Los resultados sugieren que el método evaluado mediante la implementación del protocolo establecido en la Norma Técnica Colombiana 5473 de 2007, permite evaluar la eficacia de un desinfectante bajo condiciones experimentales escogidas y controladas.


Objective. Evaluate the dilution-neutralization method proposed in the Colombian Technical Norm 5473/07, by using a gel, alcohol-based disinfectant. Materials and methods. This study was done using Pseudomonas aeruginosa ATCC 15442, Staphylococcus aureus ATCC 6538, and Enterococcus hirae ATCC 10541 as the assay microorganisms. The study was carried out at 20±1°C as obligatory temperature and additionally at 36±1°C. Four contact times between microorganisms and the disinfectant were evaluated (0, 2, 5 and 10 minutes). The assay was done both under clean conditions (0.3 g/L of bovine serum albumin), and unclean conditions (3 g/L of bovine serum albumin and 3g/L of sheep erythrocytes). Results. The implementation of this method produced precise results in all of the six repetitions used during the assay. The obtained results demonstrated a logarithmic reduction higher than five, demonstrating the bactericidal activity exerted by the disinfectant on the control microorganisms. The established experimental conditions and methodology did not affect negatively the growth of any of the strains of microorganisms. Similarly, the neutralizing used did not inhibit the development of the microorganisms of the assay. Conclusions. The method was verified by means of the fulfillment of the limits set by the rule. Our results suggest that the method evaluated by means of the implementation of the protocol established in the Colombian Technical Norm 5473/07, allows evaluating the effectiveness of a disinfectant under selected and controlled experimental conditions.


Objetivo. Avaliar o método de diluição-neutralização proposto na "Norma Técnica Colombiana 5473 de 2007", pela utilização de um desinfectante gel à base de álcool. Materiais e métodos. O teste foi realizado utilizando Pseudomonas aeruginosa ATCC 15442, Staphylococcus aureus ATCC 6538 e Enterococcus hirae ATCC 10541, como microrganismos de ensaio. As temperaturas do estudo foram de 20 ± 1°C como temperatura requerida e 36 ± 1°C e foram utilizados quatro tempos de contacto entre o desinfectante e os microorganismos avaliados (0, 2, 5 e 10 minutos). O método foi realizado sob condições limpas (0,3 g/L de albumina sérica bovina) e sujas (3g/L de albumina sérica bovina e 3g/L de eritrócitos de carneiro). Resultados. A implementação do método produz resultados precisos sobre cada uma das seis repetições realizadas no ensaio. Os resultados mostraram uma redução logarítmica superior a cinco, o que evidencia a actividade bactericida exercida pelo desinfectante contra os microrganismos control. O estabelecimento das condições experimentais e da metodologia provou não incidir negativamente no crescimento de cada uma das cepas. Similarmente, o neutralizador utilizado não inibiu o desenvolvimento dos organismos do experimento. Conclusões. Verificou-se o método através do cumprimento dos limites estabelecidos pela norma. Os resultados sugerem que o método avaliado através da aplicação do protocolo estabelecido pela "Norma Técnica Colombiana 5473 de 2007", permite avaliar a eficácia de um desinfectante em condições experimentais seleccionadas e controladas.

ABSTRACT

El proceso de esterilización de tubos anestésicos se realiza mediante una solución de glutaraldehído activado al 2 por ciento, pero el émbolo o la membrana de goma del tubo anestésico puede permitir una difusión del compuesto esterilizante. El objetivo del estudio es detectar la presencia de glutaraldehído dentro de tubos anestésicos después de aplicar protocolo de esterilización en frío (Normas de Desinfección MINSAL, 2008) mediante espectroscopía de absorción molecular. Al someter los tubos de anestésico al protocolo de esterilización podemos observar que existe una interacción entre el anestésico y la solución esterilizadora de glutaraldehído activado al 2 por ciento, entre los 220 y 250 nm, además se observa una laxitud en la membrana semipermeable después de la exposición por 10 horas al agente esterilizante. El glutaraldehído activado al 2 por ciento toma contacto con el anestésico mediante su filtración por el émbolo o diafragma.


The sterilization process is performed anesthetic tube with a solution of 2 percent activated gluteraldehyde, but the piston or diaphragm anesthetic tube allows a diffusion of sterilizing compound. The objective of this study is to detect the presence of gluteraldehyde into tubes after applying anesthetic cold sterilization protocol (Normas de Desinfección MINSAL) by molecular absorption spectroscopy. By making the pipes of anesthetic to the sterilization protocol we see that there is an interaction between the anesthetic and sterilizing solution of gluteraldehyde to 2 percent , between 220 and 250 nm, in addition there is a laxity in the semipermeable membrane after exposure for 10 hours a sterilizing agent. The activated 2 percent gluteraldehyde made contact with the anesthetic through its filtration by the piston or diaphragm.

ABSTRACT

Objetivo Determinar la frecuencia de uso de plaguicidas en la producción agropecuaria, porcícola y avícola, para comprender su influencia en el recurso hídrico en la vereda Monterredondo del municipio San Pedro de los Milagros (Antioquia) y establecer un diagnostico de su calidad. Métodos Se aplicó una encuesta domiciliaria al 100 por ciento de las familias de la vereda, sobre el uso de productos químicos y se realizaron dos análisis, el fisicoquímico y el microbiológico a tres muestras de agua en el laboratorio de Ingeniería Ambiental de la Universidad de Antioquia bajo métodos estandarizados. Resultados La quebrada El Hato presenta una contaminación por Coliformes de 1 600x103 NMP/100ml y por E.coli de 220x10 NMP/100ml. La quebrada Fray Juana presenta un grado de contaminación de 1 600x103 NMP/100ml de Coliformes. Se encontró igualmente una contaminación de las fuentes hídricas por el uso de diversos plaguicidas como: Lorsban® (Clorpirifos), Ráfaga® (Clorpirifos), Látigo® (Clorpirifos), Neguvón® (Metrifonato), Furadán® (Carbofurán). Con insecticidas como, Ganabaño® (Cibermetrina) y desinfectantes como, Límpido® (Hipoclorito de sodio 6 por ciento). Conclusión La producción agropecuaria, porcícola y avícola en la vereda, está generando impactos negativos en el recurso hídrico, por la disminución y la contaminación de éste. Las aguas de las muestras analizadas, no son aptas para el consumo de seres vivos.


Objective Determining the frequency of pesticide use in agricultural, pig and poultry production to understand its influence on water resources in the village of Monterredondo in the municipality of San Pedro de Los Milagros (Antioquia) and diagnosing its quality. Methods 100 percent of the households in the village were surveyed regarding chemical use. Physicochemical and microbiological analysis of three water samples was carried out in the Universidad de Antioquia's Environmental Engineering laboratory using standardised methods. Results The Hato stream was contaminated by 1,600x103 NMP/100ml coliforms and 220x10 NMP/100ml E. coli. The Fray Juana stream had 1,600x103 NMP/100ml coliform contamination. Water sources were also contaminated by the use of pesticides such as Lorsban (chlorpyriphs), Burst (chlorpyriphs), Whip (chlorpyriphs), Neguvon (metriphonate) and Furadan (carbophuran) and insecticides like Ganabaño (cypermethryn) and disinfectants and Limpido (6 percent sodium hypochlorite). Conclusion Agricultural, pig and poultry production in the village has been producing a negative impact on water resources due to its reduced availability and pollution. The water from the samples analysed here was not suitable for human consumption.

ABSTRACT

Práticas inadequadas de descontaminação, desinfecção e esterilização de materiais médico-hospitalares têm propiciado o surgimento de inúmeros surtos de infecções por 'Micobactérias de Crescimento Rápido' (MCR), em todo o Brasil. Entre os anos de 2000 a 2008 foram relatados mais de 2000 casos confirmados de infecções por MCR, sendo que os procedimentos por vídeo se constituíram como os maiores veiculadores destes microrganismos. O aumento do emprego de dispositivos de natureza polimérica em procedimentos médico-cirúrgicos e ausência/não cumprimento de protocolos de processamento destes materiais podem estar envolvidos na disseminação, principalmente, pela capacidade de MCR produzirem e sobreviverem em sistemas de biofilmes. Desta forma, este trabalho teve como objetivo a avaliação da susceptibilidade de cepas de Mycobacterium abscessus subsp. bolettii (suspensão e biofilmes), causadoras ou não de surto, frente a desinfetantes químicos constituídos de: Glutaraldeído 2%, Ácido Peracético 0,2%, Peróxido de Hidrogênio 35%, Solução Alcoólica de Digluconato de Clorexidina 0,5%, Solução Aquosa de Clorexidina 0,2%, Compostos de Amônio Quaternário 1,2%, Iodo 1% e Fenol 5% e Sistemas de Esterilização por Plasma (RIE e ICP) empregando mistura gasosa O2-H2O2. Paralelamente, suportes poliméricos (PVC, PEAD, PP, PUR e PC) empregados como carreadores de MCR foram analisados por Espectroscopia Fotoacústica no Infravermelho (PAS-FTIR), Microscopia Eletrônica de Varredura (MEV), microanálise em Sistema Energy Dispersive Spectroscopy (EDS) e Perfilometria. Resultados destas investigações demonstraram a resistência das cepas de M. abscessus subsp. bolettii, isolada do surto ocorrido em Belém, frente a Glutaraldeído 2%, Solução Alcoólica de Digluconato de Clorexidina 0,5%, Solução Aquosa de Clorexidina 0,2%, Compostos de Amônio Quaternário 1,2% e Iodo 1%. Numerous outbreaks of Rapid Growth of Mycobacteria (RGM) have been associated with decontamination, disinfection and sterilization malpractices, in Brazil. Between 2000-2008 were reported more than 2,000 confirmed cases due to RGM infections, and the video procedures were considered to carry these microorganisms. The increased use of medical devices in surgical procedures may be involved in the RGM spreading by its ability to grow and survive in biofilm systems. The aim of this study was evaluate the susceptibility of Mycobacterium abscessus subsp. bolletii (outbreak and nonoutbreak strains) to chemical disinfectants: Glutaraldehyde 2%, Peracetic Acid 0.2%, Hydrogen Peroxide 35%, Chlorhexidine Digluconate 0.5%, Chlorhexidine 0.2%, Iodine 1%, Quaternary Ammonium Compounds 1.2%, and Phenol 5%. Plasma Sterilization Technologies (Reactive Ion Etching and Inductively Coupled Plasma) were also evaluated. Polymers employed in medical devices (Polyvinyl chloride, High-Density Polyethylene, Polycarbonate, Polypropylene, and Polyurethane) were analyzed by Photoacoustic Infrared Spectroscopy (PAS-FTIR), Scanning Electron Microscopy (SEM), System Energy Dispersive Spectroscopy (EDS) and Profilometry. The results have demonstrated the resistance of Mycobacterium abscessus subsp. bolletii isolated from the Belém (PA) outbreak considering chemical exposition to Glutaraldehyde 2%, Chlorhexidine Digluconate 0.5%, Chlorhexidine 0.2%, Quaternary Ammonium Compounds 1.2%, and Iodine 2%. However, these strains were highly sensitive to Peracetic Acid 0.2%, Hydrogen Peroxide 35%, and Phenol 5%. The M. abscessus subsp. bolletii strains have been presented resistant to all disinfectants studied, in biofilm systems. Studies involving polymer integrity demonstrated changes in surface (oxidation and roughness) on all processed materials, and the ICP system was more aggressive in contrast to Reactive Ion Etching.

ABSTRACT

Due to the growing number of outbreaks of infection in hospital and nurseries, it becomes essential to set up a sanitation program that indicates that the appropriate chemical agent was chosen for application in the most effective way. Validating the effectiveness of decontamination and disinfection is an important and often challenging task. In order to study and compare the behavior of selected microorganisms, they were submitted to minimal inhibitory concentration (MIC). The MIC intervals, which reduced bacteria populations over 6 log10, were: 59 to 156 mg/L of quaternary ammonium compounds (QACs); 63 to 10000 mg/L of chlorhexidine; 1375 to 3250 mg/L of glutaraldehyde; 39 to 246 mg/L of formaldehyde; 43750 to 87500 mg/L of ethanol; 1250 to 6250 mg/L of iodine in polyvinyl-pyrolidone complexes, 150 to 4491 mg/L of chlorine-releasing-agents (CRAs) and 469 to 2500 mg/L of hydrogen peroxide. Chlorhexidine showed non inhibitory activity over germinating spores. A. calcoaceticus showed resistance to the majority of the agents tested, followed by E. cloacae and S. marcescens.


Devido ao número crescente de surtos de infecção hospitalar, torna-se proeminente o estabelecimento de um programa de sanitização que liste os agentes químicos a serem empregados e o modo de aplicação mais efetivo. Validação da eficácia de descontaminação é uma tarefa ao mesmo tempo importante e desafiadora. Para estudar e comparar o comportamento dos microrganismos selecionados foram realizados ensaios de concentração inibitória mínima (CIM). A CIM capaz de reduzir o bioburden inicial (>6 log10) foi: 59 - 156 mg/L de quartenários de amônia; 63 - 10000 mg/L de clorexidina, 1375 - 3250 mg/mL de glutaraldeído, 39 - 246 mg/L de formaldeído, 43750 - 87500 mg/L de etanol 1250 - 6250 mg/L de PVPI, 150 - 4491 mg/L de compostos liberadores de cloro e 469 -2500 mg/L de peróxido de hidrogênio.

OBJECTIVE: To study the bacterial contamination of antiseptics and disinfections in-use and the risk factors for contamination. MATERIAL AND METHOD: Bacterial contamination of antiseptics and disinfectants was done by culturing in-use solutions. Eight commonly used solutions were studied: alcohol 70%, chlorhexidine 4%, and 0.5%, povidone iodine 7.5% and 10%, tincture iodine 1-2%, lysol 2% and sodium hypochlorite 0.5%. RESULTS: The following risk factors for contamination were found : preparation by unskilled personnel, improper containers and prolonged use. Contamination with bacteria were found in 1.8% of 16,142 samples tested Highest rate of contamination was found in Lysol 2%. There was no contamination of povidone iodine 10% and tincture iodine 1-2%. Bacterial contamination of antiseptics and disinfectants was highest in provincial hospitals and was not found in university hospitals. The rates of contamination correlated with the duration of use. Most bacteria isolated were those found in the environment. CONCLUSION: The contamination of in-use antiseptics and disinfectants was as high as 1.8%. Risk factors for contamination were improper preparation and prolonged use.

ABSTRACT

The increasing use of alcohol as an alternative fuel to gasoline or diesel can increase emission of formaldehyde, an organic gas that is irritant to the mucous membranes. The respiratory system is the major target of air pollutants and its major defense mechanism depends on the continuous activity of the cilia and the resulting constant transportation of mucous secretion. The present study was designed to evaluate the effects of formaldehyde on the ciliated epithelium through a relative large dose range around the threshold limit value adopted by the Brazilian legislation, namely 1.6 ppm (1.25 to 5 ppm). For this purpose, the isolated frog palate preparation was used as the target of toxic injury. Four groups of frog palates were exposed to diluted Ringer solution (control, N = 8) and formaldehyde diluted in Ringer solution at three different concentrations (1.25, 2.5 and 5.0 ppm, N = 10 for each group). Mucociliary clearance and ciliary beat frequency decreased significantly in contact with formaldehyde at the concentrations of 2.5 and 5.0 ppm after 60 min of exposure (P<0.05). We conclude that relatively low concentrations of formaldehyde, which is even below the Brazilian threshold limit value, are sufficient to cause short-term mucociliary impairment

ABSTRACT

Objetivo: Avaliar desinfetantes de uso domiciliar, identificando a presença de bactérias contaminantes, e conhecer o nível de tolerância dessas bactérias ao cloreto de benzalcônio. Métodos: Foram adquiridos aleatoriamente no comércio da regiäo metropolitana de Säo Paulo, SP, Brasil, 52 amostras de desinfetantes de uso domiciliar para análise quanto à presença de bactérias contaminantes. O nível de tolerância dessas bactérias ao cloreto de benzalcônio foi determinado pelo método da macrodiluiçäo em caldo. Resultados: De 52 amostras, 16 (30,77 por cento) estavam contaminadas por bactérias Gram negativas, com contagens variando entre 104 e 106 UFC/ml. Esses contaminantes foram identificados como Alcaligenes xylosoxidans, Burkholderia cepacia e Serratia marcescens. As Concentraçöes Inibitórias Mínimas (CIM: mg/ml) do cloreto de benzalcônio para S. marcescens, A. xylosoxidans e B. cepacia foram: 2,48, 1,23 e 0,30, respectivamente. Conclusöes: Os desinfetantes de uso domiciliar a base de compostos de amônio quaternário säo passíveis de contaminaçäo por bactérias. As CIMs do cloreto de benzalcônio para as bactérias contaminantes estavam abaixo das concentraçöes do princípio ativo presente nos desinfetantes, indicando que a tolerância ao biocida näo é estável, podendo ser perdida com o cultivo das bactérias em meios de cultura sem o biocida

ABSTRACT

Nosocomial infections are an important cause of morbidity and mortality all over the world. It has been shown that appropriate enviromental hygienic and disinfection practices can be very helpful to hospital infection control. The purpose of this study was to evaluate the bactericidal activity of some disinfectants against antibiotic-susceptible and antibiotic-resistant hospital bacterial isolates. The susceptibility of 27 clinical isolates to disinfectants and antibiotics was determined by the Association of Official Analytical Chemist's (AOAC). Use-Dilution method and by the Kirby-Bauer method, respectively. All strains tested were susceptible to sodium hypochlorite, glutaraldehyde and to the association quartenary ammonium - formaldehyde - ethyl alcohol disinfectants. However, the susceptibility of strains to phenol and to one quartenary ammonium compound was variable. Among twenty-one antibiotic-multiresistant strains (methicillin-resistant staphylococci, "Enterococcus" spp, "Pseudomonas aeruginosa", "Klebsiella pneumoniae", "Proteus mirabilis", "Enterobacter cloacae", "Serratia marcescens" and "Escherichia coli") eleven (52(per cent)) and eight (38(per cent)) strains were resistant to the quaternary ammonium and phenol compounds, respectively. Among six isolates that demonstrated susceptibility to antibiotics (staphylococci, "Enterococcus" spp., "P. mirabilis", "E. cloacae" and "E. coli") two strains (33(per cent)) showed resisance to these disinfectants. The results demosntrated the lack of correlation between antibiotic-susceptibility and susceptibility to disinfectants in hospital strains

ABSTRACT

Com a crescente preocupaçäo acerca da contaminaçäo microbiana cruzada, por meio de moldes e/ou modelos, é comum a imersäo dos moldes em soluçöes desinfetantes. O objetivo deste trabalho foi verificar até que ponto estas soluçöes afetariam a capacidade de umedecimento de alginatos por gessos tipo III. Foram utilizados dois alginatos, dois gessos tipo III e duas soluçöes desinfetantes. Os moldes obtidos, quando imersos nas soluçöes, permaneceram por 10, 20 ou 30 minutos; nos respectivos modelos de gesso, mediu-se o ângulo de contato. Quanto menor este ângulo melhor era considerada a adaptaçäo do gesso ao alginato. Verificou-se que houve uma variaçäo do ângulo de contato em razäo das marcas de alginatos, dos gessos e das soluçöes desinfetantes. Houve aumento do valor do ângulo de contato à medida que se aumentou o tempo de imersäo
